What MOTS-c is

MOTS-c is a 16-amino-acid peptide with an unusual origin: rather than being encoded by the cell's main DNA, it is encoded within a short region of the mitochondrial genome (the 12S rRNA gene).1 Peptides like this are called "mitochondrial-derived peptides," and they appear to act as signals that help regulate metabolism and cellular energy.1 2 Its sequence is Met-Arg-Trp-Gln-Glu-Met-Gly-Tyr-Ile-Phe-Tyr-Pro-Arg-Lys-Leu-Arg.

MOTS-c is naturally present in the body in tiny amounts, but the synthetic version sold and studied is a laboratory-made compound. It is not an FDA-approved drug.

What the research shows - and what it doesn't

Laboratory and animal research links MOTS-c to metabolic regulation - improving insulin sensitivity and influencing weight and bone metabolism in study models.1 2 There is also some human observational data: MOTS-c levels rise with exercise, and lower levels have been associated with obesity and higher fasting insulin in some studies.1

Why MOTS-c is called 'exercise in a molecule'

MOTS-c is fascinating because it's a message from the mitochondria themselves - a signal your cells release when you exercise. In studies it improves insulin sensitivity, resists weight gain, and supports bone and muscle, and human data shows it rises with training and runs low in metabolic disease.1 2 That "exercise in a molecule" framing is irresistible - and early research partly supports it - but the large human trials that would settle it are still missing.2

MOTS-c was placed in "Category 2" of the FDA's interim list of bulk drug substances for pharmacy compounding under section 503A - the category for substances that raise significant safety concerns - and although the FDA removed it (with eleven other peptides) from Category 2 in April 2026 and referred it to the advisory committee, removal did not add it to the approved ("Category 1") list.5 6
